MORPHOLOGICAL CHARACTERIZATION OF AFLATOXIN-PRODUCING AND NON-PRODUCING STRAINS OF ASPERGILLUS FLAVUS

Abstract

Two strains- one aflatoxigenic and another non-toxigenic were grown on four culture media, namely SYMK-agar, nutrient broth-agar, sucrose-low-salt-agar and Czapek-Dox-agar, to study the effect of culture media on the colony morphology and growth of Aspergillus flavus strains. Both stains exhibited variations with respect to growth and colony morphology in different culture media. However, characters remained uniform for each culture medium used under standard growth conditions. Further, nontoxigenic strain produced dark- brown sclerotia of less than 1mm, size on Czapek-Dox-agar medium when incubated for longer duration (10-30 days) at 20°C.The development of sclerotia was studied with the help of microscopy technique, which formed due to the aggregation of specialized hyphal cells.
